Summary
Review of David Sinclair's Lifespan. Sinclair's optimistic thesis: aging is mainly epigenetic damage (the clone-starts-at-zero argument against DNA-damage theory -- a 70-year-old's DNA cloned into an embryo yields a young baby), repaired by sirtuins, which are switched on when mTOR is off (deprivation) -- hence calorie restriction / fasting / exercise / saunas / rapamycin / resveratrol / NMN as anti-aging, plus Yamanaka-factor cellular reprogramming to replace old cells with epigenetically-young ones. Scott presents the case clearly, then the skeptical counterweights: Algernon's Law (why doesn't the body already secrete the magic drug? why is the 'be weak/slow' switch there?), the mTOR power-saver tradeoff (if it makes you live longer by doing less, why aren't we in it always, and how can Sinclair claim it ALSO makes you healthier?), a friend's poke at the clone argument, and the nicotinamide null study.
